Confronto dell’utilità della subunità B8/2 ricombinante dell’antigene B, dell’antigene nativo e di un kit ELISA commerciale nella diagnosi dell’echinococcosi cistica umana
L’echinococcosi cistica (CE) è una malattia dei vermi causata dalla forma larvale di Echinococcus granulosus. Nel presente studio, la subunità B8/2 dell’antigene B (AgB) di E. granulosus è stata espressa nell’ospite di E. coli e quindi utilizzata nel kit diagnostico ELISA.
La sequenza di DNA della subunità AgB8 / 2 di E. granulosus è stata estratta da GenBank e ottimizzata per il codone. La sequenza target è stata clonata in un vettore di espressione (pGEX-4T-1). L’antigene prodotto è stato utilizzato nel sistema ELISA e la sua efficacia nella diagnosi di echinococcosi nell’uomo è stata valutata utilizzando sieri di pazienti con CE e senza CE, insieme a sieri di persone sane. Inoltre, il valore diagnostico della proteina ricombinante è stato confrontato con l’AgB nativo e con un kit commerciale.
Anticorpi contro la cisti echinococcica sono stati rilevati in 27 su 30 pazienti, corrispondenti a una sensibilità del 90% (IC 95%: 73-98%). La reazione crociata con sieri di pazienti senza CE, osservata in due casi, ha determinato una specificità del test del 93,5% (IC 95%: 82-98%) . È stata trovata una sensibilità dell’87% e una specificità del 90% per la forma nativa dell’antigene, mentre il kit ELISA commerciale aveva una sensibilità del 97% e una specificità del 95%.
I nostri dati mostrano che rEgAgB8 / 2 è una fonte di antigene adatta per la diagnosi sierologica dell’echinococcosi nell’uomo. La coespressione di rEgAgB / 2 insieme ad altre subunità AgB può potenziare l’azione di questi antigeni nella sierodiagnosi umana di CE.
L’inaffidabilità di tre kit commerciali Coxiella burnetii IgM ELISA di fase II per lo screening della febbre Q acuta umana .
La sieroprevalenza della febbre Q indotta da Coxiella burnetii (QF) è stata segnalata in varie parti dell’India. Tipicamente vengono utilizzati test sierologici/molecolari per rilevare le infezioni. Il presente studio è stato intrapreso per verificare la validità di tre diversi kit QF IgM ELISA di fase II per la diagnosi del QF acuto rispetto al gold standard Indirect Fluorescent Antibody Assay (IFA).
Cinquantotto campioni di siero di 42 pazienti (26 pazienti hanno fornito solo campioni acuti e 16 campioni sia acuti che convalescenti) che sono stati testati con tutti e tre i kit commerciali sono stati testati con IFA IgM di fase II QF per conferma.
Undici pazienti hanno dimostrato la presenza di anticorpi contro C. burnetii mediante IFA in campioni di siero acuti e/o convalescenti. Utilizzando l’IFA come riferimento, le percentuali di sensibilità, specificità, valore predittivo positivo e valore predittivo negativo per Virion-Serion / Vircell / NovaTec erano 36,36, 61,29, 25,00, 73,08; 81.82, 35.48, 31.03, 84.62 e 100, 25.81, 32.35, 100%
I tre diversi kit ELISA hanno mostrato una scarsa compatibilità tra loro e un livello inaccettabile di falsi positivi. L’IFA rimane l’unica opzione per la diagnosi di QF acuto. La discrepanza tra i risultati clinici e i risultati IFA/ELISA richiede la conferma mediante il rilevamento del DNA di C. burnetii mediante reazione a catena della polimerasi in tempo reale.
Uso clinico di un kit ELISA domiciliare per anticorpi anti-recettore della fosfolipasi A2 umana
Sviluppo e valutazione di un kit ELISA per IgG anti-PLA2R. La proteina ricombinante del recettore della fosfolipasi A2 di tipo M (PLA2R) espressa da HEK293 è stata presa come antigene di rivestimento, le IgG antiumane di coniglio marcate con HRP sono state raccolte come tag per testare le IgG anti-PLA2R mediante ELISA. Sono stati valutati l’intervallo di rilevamento lineare, l’accuratezza, la correlazione lineare, la ripetibilità e la stabilità. Inoltre, abbiamo effettuato un confronto con un kit IgG anti-PLA2R incluso. Il range di rilevamento lineare del kit non è superiore a 2 RU/ml. La deviazione relativa della precisione impostata è -15% -15% .
Esiste un coefficiente di correlazione lineare maggiore di 0,990 0 nell’intervallo 2-500 RU / ml. Il CV del test ripetibile è inferiore al 15%, quando il kit è stato posizionato a 37 ℃ a settimana, 2-8 ℃ per un anno, le prestazioni rimangono. La conformità del test al confronto nel kit anti-PLA2R IgG incluso è del 98,9% (positivo: 97,2%, negativo: 100%). Il kit Anti-PLA2R IgG ELISA da noi sviluppato è quasi identico allo standard di riferimento in cliniche specifiche e sensibili. È stato utilizzato con successo per determinare il titolo anti-PLA2R e aiutare la diagnosi clinica.
Confronto delle concentrazioni di linfopoietina stromale timica in diversi campioni biologici umani di pazienti con asma e BPCO misurati con due diversi kit ELISA .
- La linfopoietina stromale timica (TSLP) sembra essere un promettente biomarcatore dell’asma. Studi precedenti hanno esaminato principalmente i livelli sierici di TSLP. Lo scopo di questo studio era confrontare i livelli di TSLP misurati con due diversi kit ELISA nel siero, nell’espettorato indotto e nel condensato respiratorio espirato in pazienti con asma, BPCO e un gruppo di controllo.
- Lo studio ha coinvolto 24 asmatici, 36 pazienti con BPCO e 12 controlli. La concentrazione di TSLP è stata misurata utilizzando kit di ricerca e sviluppo commerciali e kit ELISA EIAab. I risultati ottenuti con il kit EIAab sono stati da 3 a 45 volte superiori a quelli misurati con il kit di ricerca e sviluppo. Differenze significative tra i gruppi studiati sono state trovate solo per la concentrazione di TSLP nell’espettorato indotto.
- Quando è stato utilizzato il kit di ricerca e sviluppo, i livelli più alti di TSLP nell’espettorato indotto sono stati trovati negli asmatici, mentre il kit EIAab ha mostrato i livelli più alti di TSLP nel gruppo di controllo. La distribuzione dei risultati nel grafico di Bland-Altman era tipica dell’errore costante proporzionale.
- La concentrazione indotta di TSP nell’espettorato può essere un biomarcatore più affidabile dell’asma rispetto al TSLP sierico. Concludiamo che il livello di TSLP dipende fortemente dal kit ELISA utilizzato per la misurazione. Pertanto, la valutazione dei risultati TSLP ottenuti con vari test può creare confusione e portare a conclusioni errate.
La determinazione della concentrazione di progesterone (P4) nel siero o nel plasma è importante per riconoscere le pecore gravide e non gravide e anche per prevedere il numero di agnelli da trasportare.
Le due metodologie più comuni per rilevare la P4 nel plasma sono il test radioimmunologico (RIA) e il test immunoenzimatico (EIA). La RIA è molto costosa e non tutti i laboratori sono adatti ad essa; EIA è disponibile in commercio per uso umano, ma solo poche aziende producono kit specifici per specie che sono costosi. Per il plasma di pecora, abbiamo verificato un kit ELISA più economico e facilmente disponibile (DiaMetra) progettato per la quantificazione di P4 nell’uomo.
Per confrontare la ripetibilità, l’accuratezza, la sensibilità e la stabilità di P4 misurata con il kit DiaMetra sono stati utilizzati pool di plasma ovino e umano. I dati sulla riproducibilità erano entro il 15% e i valori di precisione erano del 90% circa per entrambe le matrici plasmatiche. I dati di stabilità hanno mostrato una perdita <20% per congelamento-scongelamento e <30% per 30 giorni di conservazione.
